From the center of Rome, buy a metro ticket (1,50 euros) at news stands or inside the metro stations at electronic machines and take the B-line towards LAURENTINA. Get off the train at PIRAMIDE.
STAY INSIDE THE STATION and look for the signs ‘TRENI TO OSTIA’ or ‘ROMA-LIDO‘, as shown below.

The ticket that you bought before is valid 100 minutes and will include the ride to Ostia Antica since it is a neighborhood of Rome. A train leaves every 20 minutes and an electronic board will show the departure time and which platform (binario) to reach. By the way, this is the first italian DOUBLE-TRACK railway, inaugurated by Mussolini in 1924 to connect the Capital to the seaside.

Enjoy your 25 min ride to Ostia Antica Station, almost empty in the morning since most people travel to Rome to work but more crowded in the summer when romans go the beach of Ostia Lido (Lido Centro). The train goes from Porta S.Paolo towards Cristoforo Colombo (last stop).

Outside of the Ostia Antica station you will see a blue pedestrian bridge (as shown in picture below) that you need to take to cross over the road and then follow the signs to the entrance of the excavations (10 min walk) where you can buy your ticket (Adults 18 euros, EU Under 18 free, EU 18-25/students pay 2 euros). The site opens at 8.30, closing time changes according to season and is closed on Mondays. The ticket includes the visit of the medieval Castle outside and the Museum of Roman ships by the Airport of Fiumicino.
